Angiogenesis assays were performed using the V2a Kit (ZHA-4000) by TCS-Cellworks (Caltag Medsystems, Buckingham, UK) according to manufacturer's instructions. On day 1, human endothelial cells were cultured in a 24-well plate. On day 2, 24 hours after seeding, cells were treated with VEGF (positive control), suramin (negative control), and 10 µg of serum-derived exosomes (neovascular AMD, atrophic AMD, and control-derived exosomes). On days 4, 6, 8, 10, and 12 (i.e., every 48 hours) the medium was changed, and cells were retreated with the additions as described as appropriate until day 14, when tubule staining was performed, and photographs were taken. The tubule-like structures were analyzed by the TCS Cellworks AngioSys Image Analysis Software (ZHA-6000, Caltag Medsystems).
